黄海铜藻形态特征和遗传多样性研究

Study on the morphology and genetic diversity of Sargassum horneri in the Yellow Sea

  • 摘要: 为研究黄海漂浮铜藻的来源以及藻体特征,在北黄海、南黄海和青岛近岸采集漂浮铜藻,并对长岛海域定生铜藻开展调查研究。结果表明,漂浮和定生铜藻的植株形态呈现不同的季节性变化,11月两者的叶片和气囊形态差别不大,4-6月形态分化明显,表现为定生铜藻的叶片和气囊继续生长,而漂浮铜藻呈衰败趋势;漂浮铜藻的生殖托数量远低于定生铜藻,且较细短。同时,本文利用核基因(ITS2)和线粒体基因片段(cox3、rnl-atp9、rpl5-rps3和cob-cox2)研究铜藻样本的遗传多样性,结果显示,ITS2序列共发现2个多态性位点、5种基因型,线粒体基因片段共发现9个多态性位点、5个单倍型;不同地理群体间以及漂浮和定生群体间的遗传距离与群体内遗传距离差异不显著,表明该海域漂浮和定生铜藻可能来自相同的遗传群体。

     

    Abstract: In order to study the origin and characteristic of the floating Sargassum horneri in the Yellow Sea, floating S. horneri samples were collected from the northern, southern Yellow Sea and the coastal water of Qingdao. The attached samples on the aquaculture rafts along the coast of Changdao were also investigated seasonally. The study showed that the plant morphology of floating and attached S. horneri had distinct seasonal patterns. In November, there was no significant difference of the leaves and gas-filled vesicles between the floating and attached samples. Form April to June, they had obvious morphological differentiation, such as leaves and vesicles of the attached S. horneri were growing while those of the floating samples tended to decrease. The receptacles of the floating S. horneri was obviously less and slimmer than that of the attached ones. Meanwhile, one nuclear (ITS2) and 4 mitchondrial gene fragments (cox3, rnl-atp9, rpl5-rps3, cob-cox2) were selected to investigate the genetic diversity among these S. horneri samples in this study. The results showed that there were 2 polymorphic loci and 5 genotypes detected from ITS2 sequences, 9 polymorphic loci and 5 haplotypes based on the mitchondrial genes in these samples. The inter-population genetic distances among the geographic samples and between floating and attached samples were not significantly different with those intra-population genetic distances, which suggested these samples could be from the same genetic population.
